Handling Cell Line

 
Suggestion to depositor for sending cell cultures to NRFC
  • 70-80% confluent culture flask, completely filled with culture medium, supplemented with 2-5% fetal bovine serum (FBS) and properly packed with paraffin film should be sent by courier service.
Suggestion to requester for handling of cell cultures on receipt
  • On receipt of the packed culture flask, remove most of the medium, except a sufficient volume to cover the floor of the flask. Augment with 5% FBS. Incubate at optimum temperature (~28°C or as suggested) until confluency is reached.
  • Sometimes in transit, the cultures may be handled roughly and most of the cells become detached and float in the culture medium. If this has occurred, take the entire contents of the flask in the tube and centrifuge at 300g for 8 minutes. Discard the excess supernatant leaving 2 ml medium, re-suspend the cells in 5 ml of the culture medium and plant the entire cell suspension in a single 25 cm2 flask.
  • If the flask is found with turbid media or spilled over media due to broken flask during transportation, immediately inform to NRFC after receiving the flask(s) mentioning the state of flask, type of contamination, if possible, along with photo of flask and cells at email ID: nrfcnbfgr@gmail.com; basdeo.scientist@gmail.com; murali4543@gmail.com. New flasks will be sent within a month.
